Since Jesus knew in Gethsemane that God the Father's will
required him to drink from the cup, then wasn't his
request to have the cup taken away from him against
God the Father's will?

It would appear so at first. But you have to keep in mind that God is infinite, that means there would be infinite ways for God to accomplish His Will!

We are told to let our requests be known to God, yet it is always the best choice for us to say, "Not my will but yours Father be done". For the Father knows the future plans that He has for us. I have many testimony's of things that I have prayed about because I thought if was for my downfall, yet! to my suprise was turned around for my GOOD.

Phil 4:6 Be careful for nothing; but in every thing by prayer and supplication with thanksgiving let your requests be made known unto God.

However this was not the case for Jesus. He understood that He came to die on the cross. He was not looking to get out of it, he just knew that him becoming sin (for us) was shameful  and would separate him from the Father for a bit(3 days) and he was not looking forward to it.

Heb 12:2 Looking unto Jesus the author and finisher of our faith; who for the joy that was set before him endured the cross, despising the shame, and is set down at the right hand of the throne of God.

No one in his right mind would want to go through what Jesus was about to go through. He was as human as you and I.

The cup that Jesus desired to pass from Him, was NOT the crucifixion, the cup that He wanted to pass from Him, was the HOUR, His time was up, the HOUR was at hand, this is what He desired to be taken away from Him. Jesus knew full well what He was born to do, and that is to be a sacrifice for us. He was not desiring that to be passed from Him, but the HOUR to be passed, He wanted to stay longer, to be with us longer, to teach us longer, to be a shephard to His sheep longer.

Yes

Jesus had both a human nature which desired to not go through pain and suffering and His divine nature, which knew better.  In His prayer, He submits the human nature to the divine, and to His Father, giving us, forever, an example of what we also must do.
